两种小鼠乳腺癌模型的甲状腺激素和组织病理观察* Observations on Thyroid Hormone and Tissue Pathology of Breast Cancer Model in Two Species of Mice

摘要:目的:从甲状腺激素和组织病理改变探讨两种小鼠乳腺癌模型的证候(寒、热、脾虚、肾虚)属性,以期建立证病结合肿瘤模型。方法:动物均为雌性。BCML-TAⅡ99乳腺癌瘤株腋下接种于津白Ⅱ小鼠,Ca761乳腺癌瘤株腋下接种于615小鼠,均100万个/只。两种模型在不同批次于接种后第19~29天分别与正常昆明小鼠同步检测血清FT3、FT4、TT3、TT4含量和甲状腺组织病理,两种模型检测各指标时瘤重相近。并按寒证、热证、脾虚证、肾虚证评价标准评价其证候属性。结果:与昆明小鼠比较,津白Ⅱ小鼠BCML-TAⅡ99模型FT3、FT4、TT3下降,甲状腺滤泡上皮较扁平,滤泡内胶质较多;615小鼠Ca761模型FT3、FT4、TT3、TT4下降,甲状腺滤泡明显增大,滤泡上皮扁平,滤泡内胶质多。上述改变均属寒性。结论:从甲状腺激素和组织病理改变看,津白Ⅱ小鼠BCML-TAⅡ99乳腺癌模型和615小鼠Ca761乳腺癌模型可作为寒型乳腺癌证病结合模型。 Abstract: Objective: to probe into the properties of syndromes (cold, heat, spleen deficiency and kidney deficiency) in breast cancer models of two species of mice through observing the thyroid hormone and tissue pathological changes, so as to establish a model combining syndromes, diseases with tumors. Methods: The test animals are females. BCML-TA Ⅱ 99 breast cancer tumor strain was vaccinated in the armpit of TA-Ⅱ mice, and Ca761 breast cancer tumor strain in the armpit of 615 mice, about 1 million for one mouse. Two models in different batches 19 to 29 days after the inoculation received serum FT3, FT4, TT3, TT4 content detection and thyroid pathology together with normal Kunming mice. And the two models share the similar tumor weight when having the detection. The properties of syndromes were evaluated in accordance with the criteria for cold, heat, spleen deficiency and kidney deficiency syndromes. Results: Compared to Kunming mice, TA-Ⅱ mice in BCML-TA Ⅱ 99 models had FT3, FT4, TT3 declined, thyroid follicular epithelial cells flat, more glial in the follicular; 615 mice in Ca761 model had FT3, FT4, TT3, TT4 declined and thyroid follicular increased significantly, thyroid follicular epithelial cells flat, and more glial in the follicular. These changes are of the cold syndromes. Conclusion: viewing from thyroid hormone and tissue pathological changes, TA-Ⅱ mice in BCML-TA Ⅱ 99 breast cancer model and 615 mice in Ca761 breast cancer model can be regarded as the syndrome and disease combined model for cold breast cancers.
